Overview:

  • It is used in the treatment of Cancer.
  • 5-Fluorouracil is converted into fluorodeoxyuridine by Thymidine phosphorylase(TP). Fluorodeoxyuridine is then converted into fluorodeoxyuridine monophosphate which is a active metabolite. 
  • 5-FU shows its action by inhibiting DNA synthesis and repair through inhibiting thymidylate synthase(TS) which prevents conversion of deoxyuridine monophosphate into deoxythymidine monophosphate.

Anal, breast, colorkectal, oesophageal, stomach, pancreatic and skin cancers (especiallyhead and neck cancers)
